Functional identification of ygiP as a positive regulator of the ttdA-ttdB-ygjE operon.

چکیده
Functional characterization of unknown genes is currently a major task in biology. The search for gene function involves a combination of various in silico, in vitro and in vivo approaches. Available knowledge from the study of more than 21 LysR-type regulators in Escherichia coli has facilitated the classification of new members of the family. From sequence similarities and its location on the E. coli chromosome, it is suggested that ygiP encodes a lysR regulator controlling the expression of a neighbouring operon; this operon encodes the two subunits of tartrate dehydratase (TtdA, TtdB) and YgiE, an integral inner-membrane protein possibly involved in tartrate uptake. Expression of tartrate dehydratase, which converts tartrate to oxaloacetate, is required for anaerobic growth on glycerol as carbon source in the presence of tartrate. Here, it has been demonstrated that disruption of ygiP, ttdA or ygjE abolishes tartrate-dependent anaerobic growth on glycerol. It has also been shown that tartrate-dependent induction of the ttdA-ttdB-ygjE operon requires a functional YgiP.

L-Tartrate Fermentation in Escherichia coli
Escherichia coli ferments L-tartrate under anaerobic conditions in the presence of an additional electron donor to succinate. The carrier for L-tartrate uptake and succinate export and its relation to the general C4-dicarboxylate carriers DcuA, DcuB, and DcuC were studied. The secondary carrier TtdT, encoded by the ttdT (previously called ygjE) gene, is required for the uptake of L-tartrate. Th...
متن کاملA novel nucleoid protein of Escherichia coli induced under anaerobiotic growth conditions
A systematic search was performed for DNA-binding sequences of YgiP, an uncharacterized transcription factor of Escherichia coli, by using the Genomic SELEX. A total of 688 YgiP-binding loci were identified after genome-wide profiling of SELEX fragments with a high-density microarray (SELEX-chip). Gel shift and DNase-I footprinting assays indicated that YgiP binds to multiple sites along DNA pr...
متن کاملSurvey of accessory gene regulator (agr) groups and TSST-1 encoding gene (tst) in Staphylococcus aureus isolated from ewes with mastitis in the northwest of Iran
Accessory gene regulator (agr locus) is a global regulator of many virulence gene expressions inStaphylococcus aureus. Four interference classes related to genetic polymorphisms in the agr locus have sofar been described. In the present study, the agr locus specificity groups were studied within a total of 43 S.aureus isolates which were isolated from ewes with mastitis in three regions in the ...
متن کاملFunctional and Physical Consequence of Human Immunodefficiency Virus Transactivator TAT Interaction with Human Cell Cycle Regulator p53
Human immunodeficiency virus (HIV) transactivator Tat is a potent activator of both viral and cellular genes. Tat has also been implicated in the development of AIDS-related malignancy. Here, we show that Tat physically and functionally is able to sequester the cell cycle check point protein p53. This sequestration results in non-functional promoter activity of cyclin-dependent kinase/cyclin i...
متن کاملMagnetic Resonance based Sensor for the Specific Detection of Cu in Living Cells
In this study, we have developed a new contrast agent [Gd(Try-TTDA)] that can recognize Cu ion in the living cells. Moreover, the Gd complex attributes excellent selectivity for Cu over a choice of other metal ions. A gradual increases in the relaxivity and signal intensity of ex vitro and in vitro MR imaging upon Cu detection. These results implicate that a new MR based contrast agent [Gd(Try-...
